During cell growth, the medium pH can change due to metabolites released by the cells or during bacterial contamination. The presence of phenol red as a pH indicator in the culture medium allows for the monitoring of these pH variations. At acidic pH levels, phenol red turns yellow, while at basic pH levels, it becomes dark pink. For most cell culture work (at pH 7.4), the medium should be red.
